His Christian Democrats were the largest party after February's election and it was thought that a coalition with the centre-left Social Democrats would enable him to lead Germany.
His failure is unprecedented in modern times and the German parliament has 14 days to choose a chancellor.
The second largest party, the far-right Alternative for Deutschland, called for fresh elections.
