Lester is charged with first-degree assault and armed criminal action in Clay County.
He entered court with a cane, and his attorney pleaded not guilty on his behalf. His next court appearance is set for June 1.
Lester is accused of shooting Yarl last week after the teen accidentally went to the wrong house to pick up his siblings.
