הכביש מתחיל בנוף הגליל הסמוכה להר תבור וממשיך משם דרך עמק יזרעאל ועפולה לגלבוע שם נפלו שאול המלך ובנו יהונתן בקרב, ובמגידו הסמוך התעמתו צבאות ישראל עם פולשים. הכביש יורד דרומה לעיר שכם שבשומרון, אליה הגיעו לראשונה הן אברהם והן יעקב בבואם לארץ, ושם נקבר יוסף. משם לוקחת אותנו הדרך התנ"כית דרומה לשילה, בה עמד המשכן למשך 369 שנים. לאחר מכן, היא עוברת בבית-אל, המקום בו חלם יעקב על סולם מוצב ארצה וראשה מגיע השמימה. בהמשך דרומה, מגיעה הדרך ללבו של העולם – ירושלים, בירת ישראל, המקום בו מלך דוד ושני בתי המקדש עמדו על מכונם.
